We report that N-CAM, which is co-expressed with neuropilin-1 on the surface of human leukocytes, regulates leukocyte-endothelial adhesion and transendothelial migration. N-CAM is constitutively expressed on neutrophils and monocytes, but not lymphocytes, and it is not present on all human lymphoid neoplasms. In an in vitro transendothelial migration assay, monocytes from N-CAM-deficient (CD11b(+)CD18(+)) mice migrated less efficiently through an endothelial cell monolayer than did monocytes from wild-type mice. Similar results were observed in an in vivo transendothelial migration assay. Leukocyte rolling on endothelial cells in the absence of transmigration was unaffected by the absence of N-CAM. However, leukocytes from N-CAM-deficient mice rolled significantly less than did wild-type leukocytes, suggesting that N-CAM may be required for leukocyte rolling during migration. The role of N-CAM in leukocyte rolling and adhesion was confirmed in an in vitro adhesion assay in which purified human lymphocytes, granulocytes, and monocytes (from wild-type and N-CAM-deficient individuals) were perfused over a monolayer of endothelial cells in flow chambers. Monocytes and granulocytes adhered and rolled less efficiently on the N-CAM-deficient endothelial cells than on wild-type cells, as did lymphocytes, although the differences were more subtle. Neuropilin-1 appears to be the main endothelial receptor for the migration of leukocytes of both the myeloid and lymphoid lineages. The results indicate that N-CAM may have a regulatory role during leukocyte migration by mediating leukocyte adhesion and rolling, particularly during leukocyte transendothelial migration.Death row inmate charged with murder in Alabama
